Dr.-Ing. Mario A. Cypko

ME AS A SCIENTIST

As medicine becomes increasingly digital and AI-supported, the key challenge is not only to build intelligent systems, but to keep them understandable, controllable, and useful for clinicians and patients. I am interested in medical technologies that improve patient safety, decision making, and quality of life while preserving meaningful human oversight.

In my research group Medical AI and Modelling (MedAIM) at Hahn-Schickard, we focus on knowledge modelling for cancer care and transplant follow-up. Another major focus is closing the gaps between clinical snapshots through the continuous capture of patient vital signs and behaviour using smart wearables. More broadly, my work explores trustworthy clinical AI, multimodal patient representations, and Model-Guided Medicine. I am particularly interested in connecting data-driven AI with explicit, inspectable clinical models and in keeping the human-in-the-loop wherever transparency, responsibility, and meaningful clinical control matter.
 

A growing part of my work is the idea of Medical Model Science, first described by my doctoral supervisor Prof. Heinz Lemke, which I advocate as a necessary complement to Data Science and Medical Informatics. My interest here is in treating clinical models not merely as technical by-products, but as scientific and operational objects in their own right: models that need to be understandable, reviewable, maintainable, governable, and linked to clinical purpose and context. This includes questions of transparency, validation, lifecycle management, human-AI interaction, and the responsible integration of AI into real-world medicine.
 

My background is in Medical Informatics, with work on AI-based clinical decision support, multidisciplinary decision making, Bayesian and knowledge-based modelling, and human interaction with computational medical models. During my PhD at the Innovation Center Computer Assisted Surgery of Universität Leipzig, I worked on complex cancer cases and model-based decision support. I then continued as a PostDoc at the European Space Agency in the Netherlands and later at the German Heart Center in Berlin, where I explored both the capabilities and the limits of AI in demanding medical settings. Through the Excellence Cluster Matters of Activity, at the Human-Centered Computing group at Freie Universität Berlin and with the Charité Berlin, this perspective expanded further towards the question of how much AI we actually want, where we need it, and where human-centred design and the human-in-the-loop remain essential.​

At Hahn-Schickard and in collaboration with the University of Freiburg, Mario leads research in Medical AI and Modelling with a focus on trustworthy clinical AI, explicit knowledge modelling, and Model-Guided Medicine. In his research group Medical AI and Modelling (MedAIM), the main areas of work are knowledge modelling for cancer care and transplant follow-up, as well as closing the gaps between clinical snapshots through the continuous capture of patient vital signs and behaviour using smart wearables.

A central theme of his current work is the development of clinically meaningful and inspectable models that connect medical knowledge, patient context, and data-driven AI. This includes Bayesian and causal models, multimodal patient representations, and interactive clinical decision support. More recently, his work also includes LLM-guided knowledge modelling, in which large language models are used to support experts in building transparent clinical models while maintaining human oversight, validation, and model governance. He further advocates Medical Model Science as a necessary complement to Data Science and Medical Informatics, with the goal of treating clinical models as scientific and operational objects that can be understood, reviewed, maintained, and responsibly used in practice.

HUMAN-CENTERED COMPUTING GROUP

Mario joined the Human-Centered Computing group of Prof. Claudia Müller-Birn at Freie Universität Berlin and became part of the Cluster of Excellence Matters of Activity – Image Space Material. His work focused on robotic-assisted surgery as a human-centred and socio-technical setting in which surgeons, assistants, interfaces, and robotic systems continuously shape each other’s actions and decisions.

 

He studied how situation awareness, communication, and collaborative decision-making change in robotic surgery, especially under conditions where the surgeon is physically separated from the patient and the team. Based on observations in robotic-assisted surgery and later design-oriented research, his work explored how future human-robot collaboration in the operating room should be designed, how much agency surgical robots should have, and where the human-in-the-loop remains essential. This research contributed to new perspectives on human-robot interaction, interface design, and socio-material configurations in surgery.

VIRTUAL HUMAN GROUP

Focus of the project Virtual Heart Team is an AI-based system to support heart teams in therapy decision-making. 
Our latest research examines the potential outcomes of critical Type A patients. In addition, we are exploring the surgeon expertise required to achieve the desired outcome for these type A patients.

The decision support system is based on Bayesian networks and so far it seems that Bayesian networks are very good at being transparent and reliable predictors.

Biomedical Data Scientist, Human Research Office, ESTEC, ESA, Netherlands.

Focus of Mario's work was to setup a medical database system for data from ESA astronauts and ESA's simulation studies. For this, he worked closely together with his colleagues from the ESA Astronaut Center near Cologne, Germany. In addition, he had the pleasure of coordinating the scientific projects various ground-based simulation studies.

 

With the support of ESA and the International Conference on Computer Assisted Radiology and Surgery, he initiated and organized twice the conference session Digital Space Medicine together with the most experienced active ESA flight surgeon Ulrich Straube and always supported by New York radiologist Leonard Berliner, Neurosurgeon Russel J. Andrews and Medical Robotics pioneer Russel H. Taylor.

Mario joined the group Digital Patient Model (DPM) at ICCAS, Leipzig. 
He developed multi-disciplinary therapy decision support systems to support expert decisions.
The scope of his work covered new methodologies and tools for decision modelling and model interaction, using knowledge-based machine intelligent approaches, specifically Bayesian networks. The success of his work was possible through the close and open-minded team work with his clinical colleagues from the University Hospital Leipzig, specifically Dr. Matthäus Stöhr, as well as the inspiring and trustful super vision of Prof. Heinz U. Lemke, PhD and Prof. Andreas Dietz, MD.

 

Moreover, he had a research stay at the University of Pittsburgh, in USA and Polytechnika Bialystok, in Poland, under the supervision of Prof. Marek Druzdzel.

Further fruitful collaborations with Dr. Jan Wojdziak (Gesellschaft für Technische Visualistik mbH), Prof. Bernhard Preim and Dr. Steffen Oeltze-Jafra (University of Magdeburg), and many others had an essential impact on Mario's work.

Mario studied Computer Science at the Freie Universität of Berlin, Germany. During his studies he worked for ICCAS, Leipzig, as a student assistant and graduated in 2013 with his major in medical informatics and minor in psychology. In 2011, he studied abroad for one semester at the University of Limerick, in Irland. 

His diploma thesis was about Clinical Decision Support Systems, specifically diagnosing Laryngeal Cancer, under the super vision of Prof. Heinz U. Lemke.
